CHORI-321: Drosophila melanogaster (Reference Strain "PACMAN") BAC Library |

Two genomic BAC libraries ("CHORI-321" 83 kb and "CHORI-322" 21 kb average clone size) have been generated from the strain of Drosophila melanogaster used for the Reference Fly Genome Assembly. The same strain has also previously been used to create the first physical clone map using the RPCI-98 BAC library. The new "CHORI" libraries were created using the attB-P[acman]-CmR-BW vector, a special adaptation of the P[acman] vector, in combination with the E.coli strain "EPI300". Fly genomic DNA was partially-digested with MboI, size-selected by pulsed-field electrophoresis and then cloned into the BamHI site of the vector. The vector includes a conditional copy-control replicon which can be induced using commercial copy control solution (Epicenter) added to the growth media. The rationale for the libraries is explained on the PACMAN Resources website and has also been published Nature Methods, 2009 . BAC Clones containing specific genes can be identified using the search options available through the PACMAN Genome Browser . BAC clones identified this way, have the correct names recognized by the electronic clone shopping cart on the BACPAC website. For example, the BAC clone CH321-40J20 contains the "cta" fly gene. The clone name complies with the NCBI Clone Nomenclature recommendations.
