Genomic fosmid libraries were constructed for the Berkeley Drosophila Genome Project by Andreas Gnirke at Exelixis, Inc. (South San Francisco) from Drosophila stocks obtained from various sources. D. erecta and D. pseudoobscura stocks were obtained from the Drosophila species stock center, which has relocated to Tuscon (http://stockcenter.arl.arizona.edu/). The D. littoralis stock was obtained from the Umea, Sweden stock center via Kevin White (Yale). The Umea stock center is now closed. The D. willistoni stock was obtained from Jeffrey Powell (Yale). The species identifications of the stocks were confirmed by David Grimaldi (American Museum of Natural History, NY). To make the libraries, genomic DNA was partially digested with MboI, size-selected by pulsed-field-gel electrophoresis and cloned in the BamHI site of the fosmid vector pFOS1 (U.-J. Kim et al. (1992) Nucleic Acids Res. 20: 1083 -1085) in the E. coli host strain XL1-Blue MR (Stratagene).

* The DVIF01 library was previously thought to be derived from D. littoralis and was thus previously known as "DLIF01". Upon sequencing of several clones, it became apparent that a strain mixup might have occurred. Using appropiate fly genetics at the Berkeley Drosophila Genome Project Sequencing Center, the source of the library was unambiguously identified as D.virilis. To prepare the vector arms the plasmid is linearized at the AatII site, dephosphorylated and then digested with BamHI. Please note that the pUC-derived portion of the plasmid (between the two cos sites) is lost during in vitro packaging.

Individual clones identified after screening can be obtained as stab cultures at a cost of $80 per clone in addition to a $35 set-up charge per shipment. Shipping will be done by Federal Express and charged to recipients account number.
